In vessels with intact endothelium, carbachol caused hyperpolarization of the SMC membrane by 18.9±1.4 mV (10 vessels, 10 cells) and a decrease in wall tension of preconstricted mesenteric arteries by 2.6±0.2 N/m (five vessels) in normal PSS. 4-AP reduced these changes in SMC membrane potential and tension in a concentration-dependent manner (Figure 1). 4-AP 5 mM produced depolarization of SMC of resting vessels by 4.5±0.2 mV (six vessels) and attenuated carbachol-induced hyperpolarization of SMC and a tension decrease of preconstricted mesenteric arteries by 49±3% (four vessels, nine cells) and 37.7±5.3% (four vessels), respectively. 4-AP at a concentration greater than 5 mM caused an increase in wall tension of resting vessels, and an enlargement of noradrenaline contraction in normal PSS and in the presence of nitric oxide synthase and cyclooxygenase blockers.
