The polyunsaturated fatty acids (PUFA) of the n-3 series decrease the incidence of cardiovascular diseases by modifying blood properties and decreasing the occurrence of thrombosis and atherosclerosis. They also modify the membrane fatty acid composition by reducing the phospholipid n-6:n-3 PUFA ratio. Many investigators have shown that these biochemical changes in membrane phospholipid composition help decrease the severity of myocardial ischemia . This was observed in stunned myocardium after an ischemic event, with a reduction in cardiac mechanical function without a decrease in coronary flow. The main functional abnormalities in these conditions were decreased oxidative capacities and increased oxygen cost of contractility.
In a recent study, we reported that the helpful effect of n-3 PUFA in myocardial ischemia was related to energy metabolism. Compared with n-6 rich hearts, the n-3-rich hearts displayed a better postischemic recovery of pump activity associated with improved oxidative capacities and metabolic efficiency. The increased restoration of oxidative capacities was also noticed in isolated mitochondria extracted from postischemic n-3 PUFA-rich hearts. Conversely, the profitable effect of n-3 PUFA on the recovery of cardiac metabolic efficiency after ischemia was not observed in isolated mitochondria since they displayed a normal ADP:oxy-gen (ADP:O) ratio after ischemia.
