ANIMALS AND METHODS Animals and research design: Male Sprague-Dawley rats were obtained at six weeks of age (University of British Columbia Animal Care Unit) and divided into two groups: control (n=9) and fructose-hypertensive (n=9). At seven weeks of age, the fructose-hypertensive group was started on a 66% fructose diet for six weeks. The sodium content of the fructose diet was comparable with that of standard rat chow (fructose diet 4.2 g/kg, standard chow 4 g/kg). The rats were housed two per cage on a 12 h light-dark cycle and received water and food ad libitum. At the end of this period the cannu-lation procedure was carried out (described below) and direct intra-arterial blood pressure readings were recorded in response to specific intravenous drug injections, namely ACh and noradrenaline before and after L-NAME administration, and L-arginine after L-NAME administration.
Blood pressure measurement: The rats were first anesthetized with an intraperitoneal injection of pentobarbital (65 mg/kg body weight), and the right carotid artery and left jugular vein cannulated with polyethylene tubing (PE50 and PE10, respectively, Clay Adams, Parsippany, New Jersey, USA). Anesthesia was maintained with halothane gas. Mean arterial blood pressure from the artery was recorded by aDTX Plus pressure transducer system (model DT-XX, Viggo-Spectra Med, Oxnard, California, USA) linked to a Gould TA 2000 chart recorder (Gould Instruments Inc, Cleveland, Ohio, USA).
